What is Humanin?
Humanin is a 21-amino acid peptide encoded in the mitochondrial genome (16S rRNA region). It is one of the first mitochondria-derived peptides (MDPs) discovered and has potent cytoprotective, neuroprotective, and metabolic effects. Humanin levels decline with age and are inversely correlated with age-related diseases including Alzheimer's, cardiovascular disease, and diabetes.
Mechanism of Action
Humanin binds to multiple receptors including FPRL1/FPR2, gp130, and intracellular IGFBP3. It inhibits apoptosis (programmed cell death), reduces amyloid-β toxicity, activates STAT3 signaling for cell survival, improves insulin sensitivity via AMPK activation, and reduces oxidative stress in mitochondria.
Humanin Dosing Protocol
Research dosing: 2–4 mg subcutaneous 2–3 times per week. No established clinical protocol. Some researchers use 1 mg/day. Humanin analog HNG (Humanin with Gly14 substitution) is 1,000x more potent at lower doses.
