What is Kisspeptin?
Kisspeptin is a neuropeptide encoded by the KISS1 gene that acts as the master regulator of the hypothalamic-pituitary-gonadal (HPG) axis. It is the most potent known stimulator of GnRH (gonadotropin-releasing hormone) release, which drives LH and FSH production, and ultimately testosterone and estrogen synthesis. It is used in research for hypogonadism, fertility, and sexual function.
Mechanism of Action
Kisspeptin binds to the KISS1R (GPR54) receptor on GnRH neurons in the hypothalamus, triggering a powerful pulse of GnRH release. This drives LH and FSH secretion from the pituitary, stimulating testosterone production in the testes (men) and estrogen/progesterone in the ovaries (women). It is the upstream master switch of the entire reproductive hormone cascade.
Kisspeptin Dosing Protocol
Kisspeptin-10: 1–3 mcg/kg IV or subcutaneous (research doses). Kisspeptin-54: 0.24–9.6 nmol/kg IV (clinical studies). For testosterone stimulation: 100–200 mcg subcutaneous daily or pulsatile dosing. Timing matters — pulsatile dosing mimics natural GnRH pulses better than continuous infusion.
