What is Klotho?

Klotho is a transmembrane protein and circulating hormone that acts as a co-receptor for FGF23 (fibroblast growth factor 23). Named after the Greek fate goddess who spins the thread of life, Klotho levels decline dramatically with age — a 40% reduction from age 40 to 80. Low Klotho is associated with accelerated aging, cognitive decline, kidney disease, cardiovascular disease, and cancer. Increasing Klotho is one of the most promising longevity interventions.

Mechanism of Action

Klotho functions through multiple pathways: (1) as a co-receptor for FGF23, regulating phosphate and vitamin D metabolism; (2) as a circulating hormone that inhibits IGF-1 and Wnt signaling (both implicated in aging); (3) as a neuroprotective factor that reduces amyloid-β accumulation; (4) by activating antioxidant pathways (Nrf2); and (5) by modulating the TRPV5 calcium channel in kidneys.
