What is Melanotan II?
Melanotan II (MT-2) is a synthetic analog of α-melanocyte-stimulating hormone (α-MSH) that activates melanocortin receptors. It produces skin tanning without UV exposure, enhances sexual function (the precursor to PT-141), suppresses appetite, and has been used by bodybuilders for decades. It is the original melanocortin peptide from which PT-141 was derived.
Mechanism of Action
MT-2 activates MC1R (tanning), MC3R and MC4R (sexual function, appetite suppression), and MC2R (cortisol). The MC1R activation stimulates melanin production in skin. MC4R activation in the hypothalamus drives sexual arousal and reduces appetite.
Melanotan II Dosing Protocol
Tanning: 0.5–1 mg subcutaneous daily until desired tan, then 0.5 mg maintenance 2–3 times per week. Sexual function: 0.5–1 mg 45–60 minutes before activity. Start at 0.25 mg to assess tolerance (nausea is common at higher initial doses).
