What is NAD+?

NAD+ (Nicotinamide Adenine Dinucleotide) is a coenzyme found in every cell of the body, essential for energy metabolism, DNA repair, and longevity signaling. NAD+ levels decline by ~50% between ages 40 and 60, and this decline is directly linked to aging, metabolic disease, and neurodegeneration. Restoring NAD+ is one of the most evidence-backed anti-aging interventions.

Mechanism of Action

NAD+ is a substrate for sirtuins (SIRT1-7, the longevity enzymes), PARP enzymes (DNA repair), and CD38 (immune regulation). It is the central electron carrier in cellular respiration (NADH). Restoring NAD+ activates sirtuins which regulate gene expression, mitochondrial biogenesis, and cellular stress resistance.

NAD+ Dosing Protocol

IV NAD+: 250–1,000 mg IV infusion (most bioavailable). Subcutaneous: 100–200 mg/day. NMN (precursor): 500–1,000 mg/day oral. NR (precursor): 300–600 mg/day oral. For anti-aging: weekly or monthly IV infusions of 500–1,000 mg are popular.

NAD+ Benefits

Cellular energy restoration
DNA repair activation
Sirtuin activation (longevity enzymes)
Mitochondrial biogenesis
Neuroprotection
Improved insulin sensitivity
Muscle function improvement
Cognitive enhancement

Side Effect Profile

IV: flushing, nausea, chest tightness (infuse slowly). Oral precursors: generally well-tolerated. Rare: headache, fatigue. High doses may activate CD38 which consumes NAD+ (counterproductive).

Frequently Asked Questions about NAD+

Is IV NAD+ better than oral NMN?
IV NAD+ is more immediately bioavailable and produces faster, more dramatic effects (energy, clarity). NMN is more practical for daily use and produces sustained NAD+ elevation. For anti-aging protocols, many researchers use both: monthly IV infusions + daily NMN (500–1,000 mg). The IV experience is often described as transformative.
