What is Pregnenolone?
Pregnenolone is a steroid hormone synthesized from cholesterol in the mitochondria. It is the precursor to all other steroid hormones (progesterone, DHEA, testosterone, estrogen, cortisol, aldosterone) and has independent neurological effects. It is the most abundant steroid in the brain and has potent cognitive-enhancing and mood-stabilizing properties.
Mechanism of Action
Pregnenolone modulates GABA-A receptors (anxiolytic effects), activates sigma-1 receptors (neuroprotection, mood), is a positive allosteric modulator of NMDA receptors (memory), and serves as the precursor for all downstream steroid hormones. It also has anti-inflammatory effects in the brain.
Pregnenolone Dosing Protocol
10–100 mg/day oral. Start at 10–25 mg/day and titrate. Take in the morning. Higher doses (50–100 mg) are used for cognitive enhancement or hormone precursor effects. Monitor downstream hormones (DHEA, testosterone, estrogen) as pregnenolone can convert to any of them.
