What is PT-141?

PT-141 (Bremelanotide) is a synthetic analog of alpha-melanocyte-stimulating hormone (α-MSH) that acts on melanocortin receptors in the brain to directly stimulate sexual desire. Unlike PDE5 inhibitors (Viagra, Cialis) which work on blood flow, PT-141 works centrally on the nervous system to increase libido in both men and women. It is FDA-approved as Vyleesi for hypoactive sexual desire disorder in premenopausal women.

Mechanism of Action

PT-141 activates melanocortin receptors MC3R and MC4R in the hypothalamus, directly stimulating sexual arousal pathways. It increases dopamine release in the mesolimbic system, activates the paraventricular nucleus (the brain's sexual control center), and has been shown to produce erections in men with erectile dysfunction who don't respond to Viagra.

PT-141 Dosing Protocol

1–2 mg subcutaneous injection 45–60 minutes before sexual activity. Start at 0.5–1 mg to assess tolerance. Maximum: 2 mg per dose. Not for daily use — use as needed. Intranasal: 7.5–10 mg (less bioavailable).

Side Effect Profile
