What is Rapamycin?
Rapamycin (Sirolimus) is an mTOR (mechanistic target of rapamycin) inhibitor originally developed as an immunosuppressant for organ transplants. It is the only drug that has consistently extended lifespan in multiple mammalian species, including mice (up to 25% lifespan extension even when started in old age). It is used off-label by longevity researchers and physicians.
Mechanism of Action
Rapamycin inhibits mTORC1, the master regulator of cellular growth and metabolism. mTORC1 inhibition activates autophagy, reduces protein synthesis (reducing cellular 'noise'), improves mitochondrial function, reduces senescent cell accumulation, and mimics the effects of caloric restriction at the cellular level.
Rapamycin Dosing Protocol
Longevity protocol: 1–6 mg/week oral (intermittent dosing). Most longevity physicians use 2–5 mg/week or 1–2 mg/day for 2 days/week. Immunosuppressive doses (organ transplant): 2–5 mg/day — much higher than longevity doses. Blood level monitoring recommended.
