What is Semax?

Semax is a synthetic heptapeptide derived from the ACTH 4-10 fragment, developed at the Institute of Molecular Genetics of the Russian Academy of Sciences. It is approved in Russia as a nootropic drug and has been used clinically for stroke recovery, cognitive enhancement, and neuroprotection. It is considered one of the most potent nootropic peptides available.

Mechanism of Action

Semax dramatically increases BDNF (brain-derived neurotrophic factor) and NGF (nerve growth factor), enhances dopaminergic and serotonergic transmission, modulates the melanocortin system, reduces oxidative stress in neural tissue, and promotes neuroplasticity and neurogenesis.

Semax Dosing Protocol

300–600 mcg intranasal 1–2 times daily. Higher doses (1–2 mg/day) for stroke recovery or severe cognitive impairment. Cycle: 10–14 days on, 5–7 days off. N-Acetyl Semax Amidate is a more potent variant at lower doses (100–300 mcg).

Administration Routes
Intranasal (preferred)Subcutaneous injection
Half-Life
~20 minutes (but BDNF effects last 24+ hours)
