What is Spermidine?
Spermidine is a naturally occurring polyamine found in all living cells and in high concentrations in wheat germ, aged cheese, mushrooms, and legumes. It is one of the most potent known activators of autophagy — the cellular self-cleaning process. Spermidine levels decline with age, and supplementation has been shown to extend lifespan in yeast, worms, flies, and mice.
Mechanism of Action
Spermidine activates autophagy by inhibiting EP300 (a histone acetyltransferase), which leads to hypoacetylation of histones and activation of autophagy genes (ATG5, ATG7, LC3). It also has anti-inflammatory effects, improves mitochondrial function, and promotes cardiovascular health through eIF5A hypusination.
Spermidine Dosing Protocol
1–5 mg/day oral (from supplements). Wheat germ extract provides ~3 mg/g. Higher doses (10–20 mg/day) are used in some research protocols. Effects are cumulative — consistent daily use is key.
